Abstract:
In 2013, we developed a new method of microfluidic droplet manipulation based on Sequential Operation Droplet Array (SODA), which can automatically perform multi-step complex manipulation of ultra-micro (pL-nL) droplets, including formation, fusion, splitting, localization, migration and sorting of droplet. We have successfully applied SODA technology to ultra-micro-high-throughput screening, single-molecule digital PCR analysis, single-cell analysis, micro-cell experiments, and combined analysis with high-speed capillary electrophoresis, liquid chromatography, and mass spectrometry systems. Compared with the microfluidic technology reported in the literature, SODA technology has the characteristics of micro-automatic, flexibly control, versatility and wide application. It is especially suitable for multi-class, large-scale analysis and screening under the consumption of ultra-micro samples and reagents, and for performing complex, multi-step micro sample processing and analysis.
